5-Amino-1MQ, also known as 5-amino-1-methylquinolinium, is a small-molecule inhibitor of nicotinamide N-methyltransferase (NNMT). NNMT participates in nicotinamide metabolism and influences cellular pools of NAD⁺ and S-adenosylmethionine (SAM), two metabolites involved in energy metabolism and methylation processes.
Laboratory studies have shown that NNMT inhibition with 5-Amino-1MQ can increase intracellular NAD⁺ and SAM while reducing lipid accumulation in cultured adipocytes.

Much of the research on 5-Amino-1MQ has focused on body composition and metabolic function. In diet-induced obese mouse models, treatment was associated with reduced body-weight and fat-mass gain, smaller adipocytes and reduced white-adipose-tissue mass. More recent animal research also reported improved glucose tolerance and insulin sensitivity, alongside reductions in hyperinsulinemia.
Researchers have additionally investigated its effects on the liver and skeletal muscle. In obese mice, 5-Amino-1MQ reduced liver triglyceride accumulation, hepatic steatosis and inflammatory macrophage infiltration. Separate preclinical research found that NNMT inhibition increased muscle stem-cell activity following injury and improved regenerated muscle-fiber size and contractile function in aged mice.
These findings make NNMT inhibition an active research area for metabolic regulation, adipose biology, NAD⁺ metabolism, liver metabolism and muscle regeneration. However, the evidence for 5-Amino-1MQ remains predominantly preclinical, and findings from cell and animal models should not be interpreted as established effects in humans.
